Innovative high-density chip technology for counting nucleic acids 

Overview

Sample partitioning on the Clarity Plus system utilizes our proprietary chip-based partitioning technology. This innovative approach features a chip-in-a-tube design, where a high-density chip is embedded inside each tube, which is similar in size and shape to a conventional 200 µL PCR tube. The PCR mix (or "sample') is loaded onto the high-density chip with the help of a slider/platform and an auto-loader. Each sample is partitioned by the high-density chip into over 40,000 nanoliter-sized partitions. These partitions are then sealed, thermally cycled, and analyzed to identify those that are positive for PCR amplification. This allows the amount of DNA and/or RNA targets in the starting sample to be accurately quantified.

40K or 10K partitions

 

Select from two consumable formats to suit your specific needs. The 40K consumables offer higher sensitivity and a broader dynamic range, making them ideal for more demanding applications. The 10K consumables provide an accurate, reliable, and cost-effective solution for routine quantification tasks.

15 µL sample

 

Requires only 15 µL of sample, which includes your PCR mix containing primers, probes, and DNA templates. This efficient sample usage not only conserves precious samples but also delivers one of the highest resolutions on the market, with over 2,500 partitions per microliter.

8 reactions

 

Each tube strip allows for the simultaneous loading and partitioning of 8 samples in parallel, saving both time and effort. Users can choose to run between 1 and 12 tube strips, offering flexible options to accommodate varying sample throughput needs.

Tube-strips

In our chip-in-a-tube design, each tube strip closely resembles the conventional tubes commonly used in PCR experiments. This design similarity is intentional, allowing the Clarity Plus digital PCR system to integrate seamlessly with standard PCR workflows. The key distinction is that each tube contains a high-density chip. Each tube strip holds 8 of these chips, enabling the parallel analysis of multiple reactions simultaneously. All 8 reactions are loaded, partitioned, and sealed at once, significantly saving time and reducing labor. Users can choose to run up to 12 tube strips, processing reactions in multiples of 8. Since each chip generates over 40,000 partitions, a single tube strip can produce more than 320,000 data points.

Loading and partitioning

Sample loading and partitioning are automated by the Auto-Loader, with the assistance of a Slider and Platform. PCR mix is first pipetted onto the trapezoidal area of the blue Slider. The Auto-Loader then pushes the mix into the tube, where it is transferred onto the high-density chip. The mix is subsequently subdivided into over 40,000 partitions. Up to 8 samples can be loaded and partitioned simultaneously. Once the partitioning process is complete, the Slider and Platform are discarded, and the tube strip is removed from the Auto-Loader and sealed for further analysis.
